Perforamnce truning is not a single step process!

We can tune the performance of the sql server in various level

1. Server Level (os) ->check the amount of memory and cpu allocated for sql server. Use Perfmon
2.Check SQL SErver have any blocks or Deadlock. Use Activity monitor.
3.DB desing.
4.Work on Index fragmentation/ausage and statistics
5.Finally u work on your Query.

Narasimhareddy, performance can be a big topic - depending on the scale and nature of the problem.
Check here for some notes : http://www.sqlserver-dba.com/2011/06/sql_server_tuning_targets.html
Do you have some specific problem?
